Capsule Testing in Supplement Product Development: Which Capsule Types to Evaluate First

by Alejandra Rayo García 23 Oct 2026

Capsule testing is the step in supplement product development where you check how a shell behaves with your actual fill before you place a production order. It is the cheapest point to catch a problem: a brittle or softened shell costs a few bags of empty capsules to find in R&D, and a reformulation if you find it after a full production run.

Here is which capsule types to test, what to check, and how to move from prototype to production.

What Is Capsule Testing in Dietary Supplement Product Development?

In dietary supplement product development, capsule testing means treating the shell as a formulation variable, not packaging. Its material and moisture content interact with the fill and affect stability, disintegration and how the product holds up in the bottle.

At the R&D stage, the goal is to screen two or three shell materials against the same fill, confirm the capsule size holds your dose, and catch compatibility problems early. Long-term shelf life is a separate step, covered in capsule selection and formula stability.

Which Empty Capsules Should You Test First?

Which Empty Capsules Should You Test First?

Gelatin capsules are the usual baseline. They hold about 13–16% moisture and open quickly. Watch for brittleness with hygroscopic fills and softening with fills that release moisture.

HPMC vegetarian capsules run at roughly 3–7% moisture and stay flexible, so they are the first alternative for hygroscopic actives like some magnesium salts or choline bitartrate, and the default for vegan labels. They can start to disintegrate slightly later than gelatin, so test that too.

Pullulan capsules offer a stronger oxygen barrier than HPMC, which makes them worth testing for oxidation-prone powders such as CoQ10. Their water content is higher than HPMC, so skip them for very absorbent powders.

Add a specialty shell only when the formula needs it:

Create a photorealistic, premium B2B editorial image illustrating the transition from capsule prototypes to large-scale dietary supplement production and custom capsule manufacturing. Show a modern pharmaceutical and nutraceutical production environment with a stainless-steel capsule-filling machine in the background. In the foreground, display organized sample trays containing realistic empty hard-shell capsules in gelatin, HPMC vegetarian, and pullulan materials, alongside small prototype quantities and larger neatly arranged batches ready for production. Include a subtle selection of custom capsule options, such as two-tone capsules, colored shells, and capsules with tasteful printed markings, to represent custom colors and imprinting services. Keep the samples realistic and professionally arranged. The image should visually communicate how supplement brands can test small batches, confirm their specifications, and then scale to bulk orders using consistent capsule specifications. **Visual style:** Photorealistic commercial industrial photography, premium scientific editorial aesthetic, modern stainless-steel equipment, crisp capsule details, bright clean lighting, white and light gray surfaces, and subtle deep navy and blue accents. The environment should look organized, efficient, and credible for a professional supplement supplier. **Composition:** Horizontal 16:9 landscape format for a B2B blog article. Use a balanced composition with capsule samples and custom options in the foreground and production equipment softly visible in the background. Create a clear visual relationship between small-scale testing and full-scale manufacturing without making the scene cluttered. **Important restrictions:** - No text, logos, watermarks, or readable brand names. - No cartoon, illustration, or CGI appearance. - No unrealistic capsule shapes, sizes, or colors. - No finished supplement bottles, retail packaging, or cardboard shipping boxes. - Do not depict capsules as manufactured by Capsuline; focus on the testing and supply process. - Avoid exaggerated machinery, excessive props, or a staged stock-photo appearance. The final image should look like authentic professional photography for a capsule supplier's technical blog, communicating scalability, consistent specifications, custom options, and the path from prototype testing to bulk production.

How to Choose the Capsule Size for Your Prototype

The dose a capsule holds depends on its volume and your powder's tapped density. A size 0 holds about 0.68 mL, so a powder at 0.6 g/mL fills to roughly 400 mg. Need 500 mg? Move to size 00 (about 0.95 mL) or split the dose. Compare options in the empty capsule size chart.

A Simple Capsule Testing Checklist

  1. Fill the same formula into each shell, with the same size and method, so the shell is the only variable.
  2. Check fill weight and closed length. A capsule length gauge confirms capsules are fully closed, and a pill counting tray speeds up sampling.
  3. Store under two conditions: room conditions in your final packaging, and a stress condition such as 40 °C / 75% RH.
  4. Inspect at day 0, week 2 and week 4 for brittleness, softening, color change, odor and caking.
  5. Run disintegration at the start and after storage (USP <2040> covers methods for dietary supplements), and record shell lot numbers for your 21 CFR Part 111 files.
From Prototype to Production and Custom Capsule Manufacturing

From Prototype to Production and Custom Capsule Manufacturing

Capsuline sells gelatin, HPMC and pullulan empty capsules in bags of 100+ for prototype runs and boxes of 50,000+ for production, with the same quality control standards across both. You can test and scale with the same shell specification.

If you run automatic tamping-style fillers, the Tamping Simulator lets you set dosing and tamping parameters with your own formula before startup. Before ordering volume, check lot consistency, lead times and documentation, covered in buying empty capsules in bulk.

Need a branded look? Custom capsule manufacturing is the next step. Our custom capsule services cover colors, imprinting, sizes and flavors. Run one short confirmation trial on the custom shell, then request a B2B quote.
